Not absolutely sure about his "villian status," but Neil McCauley in Heat. If not the best, he's a good one.

I was all excited about this thread until I saw the no horror.... WTF!?!?

oh well, if no horror then

1.) Darth Vader (most obvious choice, you're all stupid for going else where)
2.) Hannibal the Cannibal (Border Line Horor I know but still, CLASSIC and grown up, I assume the no horror was to keep "freddy, Micheal and Jason off the thread)
3.) "The Butcher" for Gangs of New York

I can't believe one of the worst villians of all time hasnt been mentioned. Bruce Dern in "The Cowboys". He's, to this day, still known as the guy who shot John Wayne in the back.

This site is cool for this topic

http://www.afi.com/tvevents/100years/handv.aspx

they rank the top 50 villains and heroes for the last 100 years. I'm not quite positive on the criteria, and obviously they are looking to span both genres and generations, but there's some good ones on here nonetheless.

Here's a new perspective on Vader.... CLASSIC moment at the half way point when he talks about his sculpture

That evil dude from Saw (not to mention the psycho bitch) would have been awesome. I know its horror, but that's just mean.
